Technical Principle: Competitive Inhibition

The test utilizes a competitive lateral flow immunochromatographic assay format. The test strip features a specialized capture area containing an invisible Test (T) line and a Control (C) line before fluidics activation.

When the extracted seafood sample is applied, the liquid flows laterally upward via capillary action. Step-by-Step Procedure

1. Tissue Extraction & Sample Preparation

· Homogenization: Take a representative seafood tissue sample and grind or homogenize it thoroughly. Weigh exactly 1 g of the sample into a 15 mL centrifuge tube.

· Phase Separation: Add 0.8 mL of Heavy Metal Pb Reagent A and 3 mL of Heavy Metal Pb Reagent B to the tube. Cap tightly, mix/stir fully for 3 minutes, and centrifuge at 4000 rpm for 2 minutes.

· Reaction Mix (Key Step): Extract 10 µL of the clear supernatant and transfer it into a clean 2 mL centrifuge tube. Add 10 µL of Heavy Metal Pb Chelating Agent and 1.5 mL of purified water, then stir and mix well.

· Final Activation: Add 10 µL of Heavy Metal Pb Reagent C to the 2 mL tube, mix well, and the solution is fully ready for testing.

2. Running the Assay

· Open the strip container, remove the required test strips and microwells, and apply labels. (Note: Recap the strip tube immediately to shield remaining strips from humidity).

· Pipette 120 µL of the prepared sample solution directly into the microwell containing the lyophilized reagent. Mix slowly but thoroughly with the pipette tip, then incubate at room temperature for 3 minutes.

· Insert the test strip into the microwell and begin timing.

· Interpret visual lines between 5 to 8 minutes. Do not interpret after 8 minutes, as late chemical reactions are invalid.

Results Determination & Action Guide

· Negative (-): Both the Control (C) line and Test (T) line develop a strong, visible color. This confirms that the lead concentration in the seafood matrix is safely below the 0.5 mg/kg cutoff limit.

· Positive (+): The Test (T) line is visibly lighter than the Control (C) line or fails to appear completely. This indicates that the heavy metal lead contamination meets or exceeds the 0.5 mg/kg threshold.

· Invalid: The Control (C) line fails to show any visible color, regardless of the T line's appearance. The sample must be re-tested using a brand new microwell and strip.

Crucial Safety & Handling Precautions

· This kit provides qualitative detection ONLY and serves as a rapid screening tool. It is not suitable for determining exact numerical concentrations. Confirmatory analytical methods should always be used to verify positive findings.

· Never touch the exposed white reaction area in the middle of the test strip. Always handle the strip by the top end labeled 'FSTest Pb'.

· Inspect the packaging tube before use. If the tube is cracked or broken, the strips may have absorbed moisture, which can cause inaccurate results.

· All strips and microwells are strictly single-use items. Ensure assays are completed within the valid expiration period.
