Test Procedure
Step 1: Reagent Preparation — Dilute the 10× Histamine extracting solution with purified water at a 1:9 volume ratio before use.
Step 2: Sample Extraction — Mash 100g of representative sample. Weigh 1g into a 15mL centrifuge tube, add 10mL of diluted extracting solution, and shake vigorously for 3 minutes.
Step 3: Sample Dilution — Centrifuge at 4000 rpm for 1 minute. Mix 0.1mL of the supernatant with 0.9mL of histamine diluent and shake well for 30 seconds.
Step 4: Testing — Pipette 150µL of the prepared solution into a blank microwell. Insert the test strip into the microwell and read the result at 5 minutes.
Performance & Validation
The FSTest Histamine Rapid Test Strip is validated as a preliminary qualitative assay for fast screening of histamine contamination levels.
· Negative (-): Both C line and T line appear.
· Positive (+): C line appears; T line does not appear or is noticeably lighter than the control, indicating histamine presence at or above the LOD.
· Invalid: C line fails to appear, indicating a procedure or test strip failure.
