Test Procedure
Step 1: Sample Preparation — Method 1 (High Sensitivity / Concentrated)
Weigh 3.0 ± 0.1g minced sample (skin/fat removed) or shelled egg into a 15mL centrifuge tube. Add 3mL Extracting Solution A, shake vigorously for 3 min. Add 1 bag Extracting Solution B, shake for 2 min. Centrifuge at 4000 rpm for 5 min. Transfer all supernatant to a 10mL tube, evaporate to dryness with hot air at 65°C. Add 0.5mL Purifying Agent and 0.3mL Diluent Solution, vortex for 1 min, and let stand to separate. The lower layer liquid is ready for testing.
Step 2: Sample Preparation — Method 2 (Rapid Field Screening)
Weigh 1.0 ± 0.1g minced sample or shelled egg into a 15mL centrifuge tube. Add 10mL Extracting Solution C, mix up and down for 2 min, centrifuge at 4000 rpm for 3 min. Pipette 0.1mL upper supernatant into a 10mL tube, add 2mL Diluent Solution, and vortex for 1 min. The liquid is ready for testing.
Step 3: Assay Execution & Result Reading
Ensure cassette is at 20–40°C. Pipette 100µL of prepared sample solution into a microwell, mix 3–5 times, and let stand for 3 min. Transfer all mixed solution to the sample well of the test cassette. Read and interpret results after 8–10 minutes (invalid after 10 minutes).
Performance & Validation
Limits of Detection (LODs) for Target Quinolone Residues (µg/kg):

Result Interpretation:
· Negative (-): Both C (Control) line and T (Test) line appear.
· Positive (+): C line appears; T line does not appear (color intensity on T line is inversely proportional to quinolones concentration).
· Invalid: C line fails to appear, regardless of T line presence.
